Will my Call Center know that calls are being made from the web when using Push to Talk?

No, your call center will not be able to tell this information. The call will go through to the Call Center Agent as a normal phone call, and will simply answer the phone to talk to the caller. However, your company may choose to route Push to Talk calls in a specialized manner through your own internal phone system, or assign a designated phone number for Push to Talk web calls. In this case, Call Center Agents can identify that a call is coming in from the web.
