Can I 'lock' a file whilst I update it to stop my colleagues trying to update it at the same time?

No, it is not possible to lock files. However, an alternative solution to the problem is to have a single person receive all the updates and upload files to the site. If the site is large, it can be separated in several areas and have a separate uploader for each area of the site.
